'While more effective treatments are developed and someday may become more widely accessible, interventions that promote the personhood and well-being of persons living with dementia—as well as that of their care partners and caregivers—will remain integral to dementia care.'

'While Aβ is clearly the initiating factor in the context of mutations in the genes encoding APP and, likely, presenilin 1 and presenilin 2, I propose that accumulation of pathogenic forms of tau is not a consequence of Aβ in all cases of sporadic AD'

@cellpress TMEM106B is associated with brain aging; myelination disorders; and several neurodegenerative diseases, including (FTLD), (ALS), , & . Multiple single-nucleotide polymorphisms in TMEM106B have been linked to the severity of these disorders, with an association between risk alleles and increased TMEM106B expression. Different isolates can use TMEM106B for infection

'For patients in a late-stage trial, the drug called donanemab slowed progression of the disease by 35 per cent compared with those who took the placebo during an 18-month treatment period. The decline in patients’ ability to perform daily tasks was 40 per cent lower for those on the drug.

However, the drug displayed severe side effects such as brain swelling and bleeding and three people on the trial died.'

'Here’s the bottom line from it: “Mild Cognitively Impaired participants treated with anti-Aβ drugs were projected to have a material regression toward brain volumes typical of Alzheimer’s dementia ∼8 months earlier than if they were untreated.” I would say that if someone wants to make the argument that this is of no consequence (or perhaps even beneficial?) that they should step right up and have a crack at it. I (and many others) will watch with great interest. On the other hand, if the way of dealing with these findings is to pretend that they don’t exist, that is really unacceptable.'
