Hallo all - I guess an introduction might be a good idea!
I'm a mid-career (already!?) group leader, trying to unpick how different aspects of the local environment at #enhancers work together to regulate #transcription and #geneexpression.
We especially like non-coding enhancer #RNA, how their structures and dynamic movements make them functional; how disease mutations affect #eRNA activity and how the enhancer environment affects the #chromatin regulator CBP.

➡️ Our study highlights the intricate relationship between DNA methylation and enhancer regulation and reveals that most enhancers are not instructed by 5mC. Yet, methyl-sensitive enhancers are highly dependent on TET activity.
1) Minimal promoter vectors🧬🐁
#Enhancers need a minimal promoter to drive transgenes.
To test regulatory elements, we have been using the mouse beta-globin minprom.
We coupled this tiny (129 bp!) promoter in Gateway pME vectors with fluorophores, creERT2, and Gal4.
