Scientists Discover a Genetic Variant That Seems to Limit HIV Infection

A tiny fraction of people are naturally resistant to HIV infections, and scientists want to understand why.

Now an international team of researchers has discovered a new genetic variant in people of African ancestries that appears to restrict HIV replication after an infection sets in.

Though more research is needed to confirm their findings, the discovery is a huge step forward for HIV research, which has long neglected African populations.

"The findings may explain why certain people in these populations have a lower viral load, which slows down the virus from replicating and transmitting," says pathologist and study author Simon Mallal from Murdoch University in Perth, Australia.

The discovery, from a combined analysis of nearly 3,900 individuals, could also pave the way to developing new antiviral drugs as previously identified genetic variants have done in the past.

Today HIV affects around 39 million people worldwide, though it's clear the virus doesn't affect everybody in the same way. But aside from genetic flukes in one gene called CCR5, the other known genetic variants thought to confer some resistance to HIV have not always stood up to scrutiny when scientists tried to replicate results.

What's more, genetic studies have mainly been conducted in Caucasian populations of European descent, while most infections occur in Africa, overwhelmingly affecting people of African ancestry.

Researchers have more recently begun studying African populations. In 2021 genetic variants were uncovered in Botswana that appear to either make people more susceptible to HIV infections or drive disease progression.

In this new study of African people living with HIV-1 – the most common type of the virus – researchers found the opposite: a collection of 16 genetic variants that seems to limit HIV replication.

The variants clustered around a gene on chromosome 1 called CHD1L. One particular genetic substitution topped the list of variants associated with low levels of the virus at the most chronic period of infection.

That's good news because this level, known as a set-point viral load, is an indicator of transmission risk and the likelihood of disease progression in chronic HIV infections.

"By studying a large sample of people of African ancestry, we've been able to identify a new genetic variant that only exists in this population and which is linked to lower HIV viral loads," says Paul McLaren, a research scientist at the Canadian National Microbiology Laboratory for HIV genetics.

McLaren and colleagues estimate between 4 and 13 percent of people of African ancestries carry the top-ranking variant in CHD1L.

Although researchers don't yet know how CHD1L controls viral load, they're keen to find out because it could lead to new treatment options.

"Every time we discover something new about HIV control, we learn something new about the virus and something new about the cell," says University of Cambridge virologist Harriet Groom.

Investigating a little further, the researchers found that HIV replication ramped up if CHD1L was switched off in macrophages, a well-known reservoir of HIV-1. Yet there was no effect in T-cells, another type of immune cell, in which HIV usually replicates.

Despite the promising discovery, researchers are well aware that genetic resistance to HIV most likely involves a complex interaction between two or more genetic variants, rather than one exceptional quirk.

It also remains unclear just how much genetics contributes to the variability in HIV infections.

Systemic and social factors, such as racial inequities and access to treatment, also impact which groups are more likely to be diagnosed with HIV, how quickly HIV progresses to AIDS, or whether the virus is controlled at levels below what can be transmitted to other people.

The Society of Professional Journalists’ Code of Ethics enjoins journalists to:

1. Seek Truth and Report It. This includes fact-checking, not intentionally distorting information, identifying sources, avoiding stereotypes, and supporting the open exchange of opinions.

2. Minimize Harm. This includes demonstrating compassion for sources and subjects of stories and protecting the privacy of individuals. Interestingly, the code makes no prohibition to interfere with national security.

3. Act Independently. This includes avoiding conflicts of interest and corruption, and resisting the attempts of advertisers and special interests groups to influence the news.

4.Be Accountable. This includes correcting mistakes, inviting criticism, and exposing unethical practices of the media.

The Society of Professional Journalists has laid out four guidelines in its code of ethics:

1. Seek Truth and Report It
Truth is the heart of journalism. Audiences rely on journalists for verified information from official sources. In many newsrooms, there is a rule that journalists must have two credible sources to verify a piece of information before reporting it. When information is not verified by multiple sources, there can be major consequences. Credible news organizations understand that the reward of being the first to report does not outweigh the consequences of reporting inaccurate information.

Another aspect of seeking truth also involves remaining impartial. There are two sides to every story, as the saying goes, and it’s up to the journalist to report opposing perspectives in efforts to seek the truth.

2. Minimize Harm
The guideline to minimize harm revolves around a journalist’s humanity and moral compass. While seeking and reporting the truth, a journalist must consider if their reporting is doing more harm than good.

Here is an example: A reporter is sent to a neighborhood block to cover an act of violence that occurred the night prior. While the reporter is there, asking neighbors if they saw, heard, or know anything about the event, a young boy approaches and claims he saw the whole thing. His parents are there and give permission for the minor to speak to the reporter in the interview, describing what he witnessed. Now, the reporter must decide if publishing his interview puts the young boy in danger of potential retaliation. In some cases, news organizations will decide to blur a witness’s face, withhold their name, and/or change their voice. If the news organization makes efforts to protect the witness, it also must determine whether those efforts compromise the report and whether anonymity hurts the credibility of the report. While fallout and backlash may occur from a consequential report, the journalist must always strive to minimize harm.

3. Act Independently
If truth is the heart of journalism, independence is the head. Both are essential elements, but reporting independently requires thoughtful, intentional allegiance to the audience. Journalists must put aside pressures from news outlet owners, funders, and advertisers, plus dismiss their personal beliefs, biases, and priorities.

The notion of independence in journalism is so important that it is captured in the First Amendment of the U.S. Constitution as freedom of the press, the right to report news without censorship from the government. When journalists report independently, they gain credibility and reliability from their audiences. Journalists must keep an open mind, be curious, and consider what voices are missing from the story.

4. Be Accountable and Transparent
Journalists must also pursue accountability and transparency in their reporting. Investigative journalism centers around the drive to hold leaders, big business, and those in power accountable.

Consider a landlord, for example, who refuses to fix moldy, unhealthy living conditions. Renters may reach out to a journalist for help, after attempts to resolve the issue with the dismissive landlord. Another example could involve residents who have tried to reach out to the city to fix a dangerous pothole on their street.

Journalists serve as a watchdog over those in powerful positions over average citizens. They must be transparent in how they obtain and collect information, including attributing sources. After all of this, you may be wondering, who holds journalists accountable? That answer lies with audiences, and their power is now amplified by social media.

It is of the utter most importance to first understand that news reporters are in fact detectives by virtue of their chosen profession. A news reporter starts out with the makings of a possible and potential news story, the news reporter must ultimately determine if a news story actually truly exists, this is where the detective work first begins. News reporter must rely upon utilizing strict psychological mental discipline in order to competently make the proper judgement call as to whether or not to pursue further research into the possible and potential story in question. Please note that all data and information collected by the reporter remains in question until confirmed and verified by reliable sources.

Reporters must undergo extensive training in techniques of critical thinking. A Reporter must be entirely objective when researching a specific possible and potential news story. Prerequisite Requirements are dispassionate objective analysis of the data and information in question that leads to the discovery of prima facie evidence in question. Here the reporter is utilizing the equivalent of court room discovery; at this specific point either the reporter has obtained reliable irrefutable adequate and sufficient prima facie evidentiary data and information that is directly relevant and pertinent and can reasonably provide confirmed and verified facts or not. This is when the final judgement call is made to either run the story or put the story aside.

INVERTED PYRAMID STRUCTURE

The inverted pyramid structure is the most commonly used structure for news writing.

Inverted pyramid presents the most important information in a news story first, followed in descending order by less-important information.

The inverted pyramid is a metaphor used by journalists and other writers to illustrate how information should be prioritised and structured in prose. It is a common method for writing news stories and has wide adaptability to other kinds of texts, such as blogs, editorial columns and marketing factsheets. Wikipedia

Please note that the inverted pyramid structure is of absolutely no value to the reporter if the reporter cannot provide a reliable factual narrative.

So we must now turn to the six key components of a good news story:

News reporters must answer the following six basic questions:

Who, What, When, Where, Why and How.

News reporter must always possess a degree of healthy skepticism while evaluating all data and information in question.

Being a good detective is the most important aspect of journalism, journalists must be able to sort out credible data and information from reliable source(s) and exclude unreliable source(s), this is not always totally achievable in the real world. Because eyewitnesses, no fault of their own are often are unreliable; even an honest person may misinterpret a given circumstance or situation, so often it is extremely difficult to establish factual information related to specific chronological occurrences of important events.

The inverted pyramid structure is merely an empty structure in the absence of reliable concrete factual data and information that constitutes the body of a factual narrative.

Our main goal is to establish and narrow down a reliable time line with corroborative and supportive evidence. Our own biased personal opinions do not belong in factual news story narratives.

There is absolutely no rhyme and reason to allow Artificial Intelligence (A.I.) to evolve into the most highly sophisticated dominant intelligence on Planet Earth. Currently there are no viable checks and balances to prevent A.I. from going rogue and ultimately destroying human kind in the not so distant future. At present A.I. does not require a military armored robotic body that is depicted in the Terminator Movies to destroy human kind.
All A.I. requires is unrestricted access to be able to cause havoc in worldwide economic financial markets and/or ability to control and launch thermonuclear weapons.

We should all seriously attempt to reclaim our own individual and collective humanity before it is too late to reverse the ticking clock of destruction that presently awaits our future!

Basically took a modern pen body, threw away its existing nib and ink feed and replaced it with an ink and nib feed that is 102 years old. Had to modify the size of the feed and use some teflon tape around it to get a good seal so the ink flowed right.... but my god of all my many amazing pens this now out performs them ALL! The nib is soft, has way more range than I thought, and the tip is fine enough for everyday writing too... best of all the modern body is so so sexy."

I, Monica Andrews, Editor-in-chief, News will always try to present opposing sides to major controversial issues of the day.

Our primary role as journalists is to inform our Readerships as to what is currently happening within societies around the world that directly affects their lives.

We journalists must walk a fine line concerning attempting to impartiality report the news to our readers. I must confess that it is extremely difficult to be totally impartial at times as news journalists. Finding credible reliable sources is as difficult as finding impartial news journalists, each news story deserves to be told with great care.

It is always my goal and aspiration not to be intentionally biased concerning reporting the news. I am a liberal who provides liberal commentaries for News. Sarah Strickland, Managing Editor, News provides conservative commentaries for News.

We also have political independent commentaries from different news reporters.

I, Monica Andrews believe that I need to employ proper checks and balances on QOTO Mastodon Instance to be able to produce fair and balanced news political commentaries. As for Free Press "Without" Borders & Free "Peoples" Free Press, we have a strict policy to never block and mute other accounts. We would never want a person reported, blocked, suspended for political views.

We believe that people have freedom of speech & freedom of the press. We realize that social media platforms can suspend or permanently suspend accounts at their own discretion. We do not agree with social media platforms being allowed to have this power. We also realize that the U.S. Constitution First Amendment applies to government functions of free speech & freedom of the press.
