Looking for suggestions: I'm trying to remove a Boc in the presence of an acetamide. I usually just whale on the Boc, but those conditions might remove the acetamide (in related systems I've cleaved the acetamide in conc. HCl at 50C for 35 min). Do I risk concentrated HCl at room temp? Do I try TFA?

Random stuff found during minor lab rearrangement: unopened bottles of Superdex 30 & 200. Ancient, but probably still good. It's not like I plan to freakin' purify peptides by hand any time soon, but it's way too expensive to throw away. *sigh*

But it's a long weekend (I was wondering why it seemed to empty out earlier than usual) and I am done. Into the freezer it goes.

One of the great mysteries: I put a larger flask on the CH2Cl2 outlet for the solvent system, thinking it still wasn't going to pull solvent (it hasn't worked in ~1 year). Started filling instantly.
If it was actually back siphoned it should have taken time to fill, so that wasn't it. Maybe it wasn't working because sometimes I have the system closer 6 psi rather than 9 psi? Could it make that big a difference for dispensing the heavier solvent?
I have no idea!

If you are an undergrad in a lab and you need something, do not start looking through random drawers that don't belong to your lab. Particularly if the person they do belong to is literally standing right there.
I would have given you a glass pipet if you asked, but because you acted like a jerk (no "sorry I didn't know") I told you to go ask someone from your lab to get you one.
Relatedly: don't borrow equipment without asking (this one also applies to non-undergrads).
We have a planned power outage on Sunday that will necessitate generators being moved on-site for essential equipment. In order to reduce the load I have to unplug everything that isn't needed over the weekend. You don't realize how much stuff just sits in standby in a lab until you have to track it all down...

My bachelor student and I performed some TGA #measurements yesterday, using these cute, tiny crucibles.
The ceramic "plates" the samples (5-15 mg) are put on are smaller than the tip of my pinky.
The sample holder is picked up with a hook which is attached to an extremely sensitive scale and then put into an oven which heats the samples to 1100 °C, while continuously monitoring the weight of the sample to see exactly when different organic products start to evaporate.

I have a reaction with two starting materials that could both be considered "precious". I made an estimate of the amount of A (B was much more precise) and it looks like A has not been fully consumed. I think B has been, but B seems to get consumed even when things go wrong.
Do I add more B (plus additional reagents, which were scaled to the estimated amount of A)?
Recovering either after the reaction is very difficult/impossible.

Nothing like a day with a water bath extraction...
The theoretical yield from the giant column was 7.6 grams, I got maybe 0.3 from that giant column and there didn't seem to be a huge amount of other stuff (I was expecting to recover a lot of starting material).
I knew it volcano'ed earlier (since I was the fool who caused it), but I thought I only lost a small amount. Good thing I kept that bath...though now I'll have to rerun the column.

When you have to scale up your chemistry. To make life more fun the compound is not UV active...
Unfortunately the yield looks like it's going to be much worse, not sure if it was the solvent ratio change or the slight decrease in excess reagent on this scale.
